This experimental study was carried out to examine the effect of isoniazid on the glycogen and glucose-6-phosphatase in mice liver. The animals which were orally administered with a single dose of 0.5mg isoniazid per individuals were sacrificed as different time lapses.
The one part of the removed liver was fixed in absolute alcohol and stained with the Best¢¥s carmine for glycogen.
The another was frozen-sectioned by cryostat for glucose-6-phosphatase.
The results were as follows;
1. The contents of glycogen began to be decreased from the first day and was markly fallen at the sixth and seventh day.
2. The glucose-6-phosphatase activities began to be decreased from the second day and appeared to be markly diminished at the sixth, and at the seventh, the activity of the central area is negligible.
3. The change of glycogen contents and glucose-6-phosphatase activities were to be decreased from the central area. The decrease of the glycogen contents was marked at the cytoplasm of liver cells and the fall of the glucose-6-phosphatase activities was marked at the intranuclear and perinuclear region of liver cells.
4. Histopathologic changes were lymphocyte infiltration, local hepatocellular vacuolation, ballooning degeneration, and mild lobular disarray.
